Project: Spatial and functional mapping of Polo like kinase 1 (Plk1) at the centromere-kinetochore axis

Polo like kinase 1 (Plk1) is a master regulator of mitosis and is involved in maintaining chromosome segregation fidelity and structural integrity of the centromere-kinetochore architecture. I am interested in spatially mapping Plk1 and identifying its substrates and/or binding partners at the centromere-kinetochore axis (~100 nm in length). To map the distributions of Plk1 along this axis, I’m developing a modified Expansion Microscopy method which provides a resolution of ~15 nm. To identify the binding partners/substrates, I’m using a proximity based labelling approach combined with Mass Spectrometry.
